Question Regarding “Transient Expression of Proteins Using COS Cells”
In the protocol describing transient expression of proteins in COS cells (CPMB Unit 16.12) isn’t it desirable to wash cells after aspirating the DMEM/DEAE-dextran/DNA step (Basic Protocol, step 5) and to wash after aspirating the DMSO (Basic Protocol, step 6)?

Some investigators do in fact add these washing steps to the procedure and find that they have some impact on the efficiency of transfection. The authors do not use the washes and did not include them in the protocol because they add too many steps to the procedure and their value was found to be marginal.
